To calculate the potency of a substance using HPLC on an "as is" basis, you'll need to determine the ratio of the sample's peak area to the standard's peak area, then account for their concentrations and the standard's potency. The formula is: 

Potency (As is) = (Sample Area / Standard Area) * (Standard Concentration / Sample Concentration) * Standard Potency
