Difference between UPLC & HPLC
Will there be any difference in selection of the columns between these two methods and which will elute the compound faster?

As compared to HPLC, UPLC will be used with short columns
So, UPLC has faster elution
Columns for HPLC is normally of 150mm in length where as UPLC is of 50-75 mm in length
And also, micron size and pore size of UPLC columns are less than HPLC
So, for faster elution, better use UPLC

If LOQ is not given in the Method validation Protocol, we have to establish the LOQ Level and then include it in the validation.
If you are a Quality control analyst, we should think of you as such
So, if we go deep into the topic, before preparing a validation protocol, the method will be checked for its feasibility. In feasibility, you could check for LOQ level for the establishment.
Establishment of LOQ level shall be done by using various methods (three methods) and here mostly, we use the signal-to-noise ratio method.
You should prepare your solution that can go as low as lower and s/n is around 10.
Speaking generally, we could achieve that with having our LOQ concentration solution with 5000-7000 Area.
If we are not getting enough s/n with 5000-7000, we could go higher, but make sure that the LOQ of choosing concentration is as high as possible to 10% w.r.t. the Standard 100% Level in the Linearity level.
Once we achieve that, we could establish and check for LOQ Precision.
